This medicine may be recommended for different uses. Approach your physician or pharmacist for more information.

Do not use Ofladon in people with the following conditions:

  • Hypersensitivity
  • Pregnancy and breastfeeding: Nintedanib can cause harm to a developing fetus, and therefore, it is contraindicated during pregnancy. It is important to use effective contraception during treatment with nintedanib and for at least three months after the last dose. Additionally, the use of nintedanib is not recommended during breastfeeding, as it is not known if the medication is excreted in human breast milk.
  • Severe liver impairment: Nintedanib is primarily metabolized in the liver, and individuals with severe liver impairment may have difficulty metabolizing the medication. Therefore, nintedanib is contraindicated in patients with severe liver impairment. Your healthcare provider will assess your liver function before starting nintedanib to ensure it is safe for you.
  • Severe renal impairment: Nintedanib is primarily excreted through the kidneys, and individuals with severe renal impairment may have difficulty eliminating the medication from their bodies. Therefore, nintedanib is contraindicated in patients with severe renal impairment. Your healthcare provider will assess your kidney function before starting nintedanib to ensure it is safe for you.

It is important to discuss your medical history and any pre-existing conditions with your healthcare provider before starting nintedanib. They will assess the potential risks and benefits and determine if nintedanib is appropriate for your specific situation.

Ofladon may have the following side effects:

  • Gastrointestinal issues: Nintedanib can cause gastrointestinal side effects such as diarrhea, nausea, vomiting, abdominal pain, and decreased appetite. It is important to stay hydrated and maintain a balanced diet while taking nintedanib. Your healthcare provider may recommend medications to manage these side effects.
  • Liver toxicity: Nintedanib may cause liver toxicity, which can manifest as elevated liver enzyme levels in blood tests. Symptoms of liver toxicity may include yellowing of the skin or eyes (jaundice), dark urine, and abdominal pain. Regular monitoring of liver function is essential during treatment with nintedanib.
  • Bleeding: Nintedanib can increase the risk of bleeding. This may manifest as nosebleeds, blood in the urine or stools, or prolonged bleeding after minor cuts or injuries. It is important to inform your healthcare provider if you experience any signs of bleeding.
  • Cardiovascular effects: Nintedanib may cause an increase in blood pressure and heart rate. Regular monitoring of blood pressure is important during treatment. In some cases, nintedanib may also increase the risk of blood clots, so it is important to be aware of any signs of blood clotting, such as leg pain or swelling, chest pain, or shortness of breath.
  • Skin reactions: Nintedanib may cause skin reactions such as rash or itching. If you experience any skin changes, it is important to inform your healthcare provider.

Precautions

Take extra caution in patients with the following medical conditions:

  • Bleeding risk: Nintedanib can increase the risk of bleeding. If you have a bleeding disorder or are taking medications that increase the risk of bleeding, such as anticoagulants, it is important to inform your healthcare provider. They will assess the benefits and risks of nintedanib treatment in your specific situation.
  • Pregnancy and breastfeeding: As mentioned earlier, nintedanib can cause harm to a developing fetus and is contraindicated during pregnancy. If you become pregnant while taking nintedanib, it is important to inform your healthcare provider immediately. Nintedanib is also not recommended during breastfeeding, as it is not known if the medication is excreted in human breast milk.
  • Liver and kidney function: Nintedanib is processed mainly by the liver and eliminated through the kidneys. Your healthcare provider will assess your liver and kidney function before starting nintedanib to ensure it is safe for you. Regular monitoring of liver and kidney function may be necessary during treatment.
  • Lung conditions: Nintedanib is primarily used to treat certain lung conditions, such as idiopathic pulmonary fibrosis. However, it is important to discuss your specific lung condition with your healthcare provider to determine if nintedanib is the appropriate treatment option for you.

Prior to taking this medicine, tell your physician or pharmacist if you are allergic to it, or if you have some other hypersensitivities. This drug may contain inert ingredients which can cause unfavorable side effects or different issues. Talk with your pharmacist for more information.

Interactions

Ofladon may interact with the following medicines:

  • Strong CYP3A4 inhibitors: Medications that are strong CYP3A4 inhibitors, such as ketoconazole, clarithromycin, and ritonavir, may increase the levels of nintedanib in your body. This can potentially increase the risk of side effects. Your healthcare provider may need to adjust the dose of nintedanib if you are taking such medications.
  • Strong CYP3A4 inducers: On the other hand, medications that are strong CYP3A4 inducers, such as rifampin, phenytoin, and St. John’s wort, may decrease the levels of nintedanib in your body. This can reduce the effectiveness of nintedanib. Your healthcare provider may consider alternative treatments if you are taking such medications.
  • Anticoagulants: Nintedanib may increase the risk of bleeding, and if you are taking anticoagulant medications, such as warfarin or aspirin, the risk of bleeding may be further increased. It is important to inform your healthcare provider if you are taking any anticoagulants, as they will monitor your condition closely and adjust your treatment plan accordingly.
  • Proton pump inhibitors (PPIs): PPIs, such as omeprazole and esomeprazole, can reduce the absorption of nintedanib. If you are taking a PPI, your healthcare provider may consider alternative medications to manage acid-related symptoms.
  • Other medications: Other medications, such as certain antibiotics, antifungals, and heart medications, may have interactions with nintedanib. It is important to disclose all your medications to your healthcare provider for a comprehensive evaluation of potential interactions.

Inform your physician about the drugs you take including over-the-counter medications, prescribed drugs, supplements and vitamins.

This is not a complete list of drug interactions. Approach your physician or pharmacist for more details.

The effects of certain medications can change if you consume different medications or herbal products simultaneously. This can build your risk for side effects or may cause your drugs not to work effectively. These interactions are conceivable, yet don’t generally happen. Your physician or pharmacist can prevent or oversee interactions by changing how you utilize your prescriptions or by close checking.

While utilizing this drug, don’t start, stop, or change the dose of other medicines you are using without your physician’s advice.

If you suspect an overdose, contact your doctor right away.
