DEMECLOCYCLINE

Demeclocycline is a drug is known as an antibiotic. It is said to inhibit protein synthesis in bacterial cells by binding with the 30S and possibly the 50S ribosomes. It acts on the cell membrane of bacteria and may also inhibit the action of antidiuretic hormone (ADH) in patients with chronic syndrome of inappropriate antidiuretic hormone secretion (SIADH).

Demeclocycline is used to treat infections such as atypical pneumonia. It is also used to treat chronic hyponatraemia associated with SIADH

Demeclocycline should not be given to children below 12 years of age. It should also not be given to people with allergy to demeclocycline or other tetracyclines, as well as those who are breastfeeding and those who are using methoxyflurane.

Demeclocycline may give rise to the following side effects:

  • Photosensitivity
  • Reversible nephrogenic diabetes insipidus
  • Permanent discolouration of the teeth
  • Brownish-black microscopic discolouration of thyroid tissue
  • Nausea
  • Vomiting
  • Diarrhoea
  • Increases in liver function test values
  • Hepatitis
  • Jaundice
  • Liver failure
  • Rashes
  • Pruritus
  • Bullous dermatoses
  • Exfoliative dermatitis
  • Skin discolouration
  • Pseudotumour cerebri in adults
  • Bulging fontanelles in infants
  • Headache
  • Dizziness
  • Visual disturbances
  • Impaired hearing
  • Urticaria
  • Angioneurotic edema
  • Pericarditis
  • Anaemia
  • Thrombocytopenia
  • Neutropenia
  • Agranulocytosis
  • Eosinophilia
  • Anaphylaxis
  • Stevens-Johnson syndrome
  • Clostridium difficile-associated disease

Precautions

Extra caution should be given in people with the following medical conditions:

  • Patients with pre-existing SLE and myasthenia gravis
  • Liver and kidney impairment
  • Pregnancy

Demeclocycline can interact with the following medicines:

  • Antacids containing Aluminum, Magnesium, or Calcium
  • Iron-containing preparations
  • Zinc
  • Kaolin
  • Quinapril
  • Strontium ranelate
  • Sucralfate
  • Tripotassium dicitratobismuthate
  • Oral contraceptives
  • Penicillins
  • Retinoids (e.g. acitretin, isotretinoin, tretinoin)
  • Ergotamine
  • Methysergide
  • Oral typhoid vaccine
  • Anticoagulants
  • Methoxyflurane
