Tegafur is a 5-fluorouracil (5-FU) prodrug that is metabolized to 5-fluoro-deoxyuridine-monophosphate, which is the active metabolite (FdUMP). Thymidylate synthase and DNA synthesis are both inhibited by FdUMP. Uracil inhibits dihydropyrimidine dehydrogenase (DPD), the main enzyme that metabolizes 5-FU, raising 5-FU exposure and operation.
