Perindopril, a prodrug of perindoprilat, is an ACE inhibitor. It stops angiotensin I from being changed into angiotensin II. This increases the activity of renin in the plasma and decreases the production of aldosterone and substances that narrow blood vessels.

Amlodipine, a dihydropyridine Ca channel blocker, prevents Ca ion transmembrane input into cardiac muscle and vascular smooth muscle, resulting in vasodilation and relaxation. It also works directly on vascular smooth muscle, lowering blood pressure and peripheral vascular resistance.
