Pembrolizumab is a humanized immunoglobulin G4 monoclonal antibody that binds to the negative immunoregulatory protein programmed death-1 (PD-1), preventing it from interacting with ligands PD-L1 and PD-L2. When the PD-1 pathway is blocked, tumor-specific cytotoxic T-lymphocytes are reactivated, and an immune response to tumor cells is induced. It is also known as lambrolizumab.
