Description: Omeprazole is a Proton Pump Inhibitor (PPI) that blocks gastric acid secretion.

Onset: Approx 1 hr.

Duration: Up to 72 hr.

Pharmacokinetics:

Absorption: Rapid but variably absorbed (oral). Bioavailability: Oral: Approx 30-40%. Time to peak plasma concentration: Approx 1-2 hr.

Distribution: Plasma protein binding: Approx 95%.

Metabolism: Hepatic metabolism via CYP2C19 isoenzyme to form hydroxyl-omeprazole and CYP3A4 to form omeprazole sulfone.

Excretion: Mainly via urine (approx 77%), the remainder in faeces (via the bile). Elimination half-life: 0.5-3 hr.

  • Used to treat acid reflux and ulcers by decreasing the amount of acid produced by the stomach
  • Relieves symptoms such as heartburn, difficulty swallowing, and persistent cough
  • Aids in healing acid damage to the stomach and esophagus
  • Ulcer prevention
  • May aid in preventing Esophageal Cancer
  • It is a proton pump inhibitor (PPI)

 

 

How to use omeprazole

  • Always refer to your doctors instructions before taking this medication. Duration and dosage will be determined by your medical condition and its response to the medication. Make sure to take this mediciation in the prescribed amount and time period even after symptoms disappear. Discontinuing the medication before completion of treatment may cause incomplete treatment of condition. If Symptoms persist, consult your doctor.
  • Taken by mouth; Before a meal; usually once a day; swallow capsule whole
  • If you are also taking sucralfate, take omeprazole at least 30 minutes before
    • Minor side effects (if these symptoms persist or worsen, inform your doctor)
      • Headache
      • Abdominal pain may occur. If any of these effects persist or worsen, tell your doctor or pharmacist promptly.

     

    • Serious side effects (inform your doctor immediately)
      • Low magnesium blood level (such as unusually fast/slow/irregular heartbeat, persistent muscle spasms, seizures)

     

    • May rarely cause a severe intestinal condition (Clostridium difficile-associated diarrhea) due to a type of bacteria.
      • Inform your physician immediately if you develop: persistent diarrhea, abdominal or stomach pain/cramping, fever, blood/mucus in your stool.

     

    • Rarely have caused vitamin B-12 deficiency
      • Risk is increased if they are taken every day for 3 years or longer
      • Inform your physician immediately if you develop the following symptoms: unusual weakness, sore tongue, or numbness/tingling of the hands/feet

     

    • Severe allergic reactions are rare. Get immediate medical help if you notice the following:

    Rash, itching/swelling (especially of the face/tongue/throat), severe dizziness, trouble breathing kidney problems (such as change in the amount of urine)

    • Precautions:
      • Inform your doctor if you
        • Are allergic to the medication or to similar drugs such as esomeprazole, lansoprazole, pantoprazole.
        • Have a history of liver disease
        • Get immediate medical help if you have:
          • Heartburn with lightheadedness/sweating/dizziness, chest/jaw/arm/shoulder pain (especially with shortness of breath, unusual sweating), unexplained weight loss.
          • Trouble/pain swallowing food, bloody vomit, vomit that looks like coffee grounds, bloody/black stools, heartburn for over 3 months, frequent chest pain, frequent wheezing (especially with heartburn), nausea/vomiting, stomach pain

       

      • May increase risk for bone fractures with longer use, higher doses, and in older adults
        • Discuss with your doctor about method in bone loss/fracture prevention
        • Calcium and Vitamin D Supplements

       

      • Children may be more sensitive to the side effects of this drug, especially fever, cough, and infections of the nose/throat/airways.

       

      • During pregnancy, discuss the risks and benefits of use with your doctor.
      • Consult your doctor before breast-feeding as this medication passes into breast milk and may cause unwanted effects on your baby

       

      Interactions:

      Taking multiple drugs may affect how medications work and/or cause unwanted side effects. List down all the medications you are taking and inform your physician and pharmacist. Do not take, discontinue, or alter any dose of medications without prior approval of your physician.

       

      • Products that may interact with this medication Include:
        • Cilostazol
        • Clopidogrel
        • Methotrexate (especially high-dose treatment)
        • Rifampin
        • St John’s wort.

       

      • This medication decreases stomach acid that may affect the following products:
        • Atazanavir
        • Erlotinib
        • Nelfinavir
        • Pazopanib
        • Rilpivirine

      Certain azole antifungals (itraconazole, ketoconazole, posaconazole)

    • Overdose:If overdose is suspected, contact a poison control center or emergency room immediately.Missed Dose

      If you miss a dose, take it as soon as you remember. If it is near the time of the next dose, skip the missed dose and As soon as you remember, take a regular dose or if the time of the next dose is near, wait for it before taking another dose and resume your usual schedule. Do not increase the dose to compensate for the missed dose.
