FIDAXOMICIN

Fidaxomicin is a drug that belongs to a class of medicines known as macrocyclic antibiotics. It inhibits the bacterium Clostridium difficile through inhibition of RNA synthesis within its cells by binding to RNA polymerase sigma subunit. It is usually given by the mouth.

Fidaxomicin is used in the treatment of diarrhoea and colitis caused by the bacterium Clostridium difficile.

Do not give to people with known hypersensitivity to Fidaxomicin.

Fidaxomicin can give rise to the following side effects:

  • Hypersensitivity reactions (e.g. dyspnoea, pruritus, rash, angioedema of mouth, face and throat)
  • Dizziness
  • Headache
  • Nausea
  • Gastrointestinal haemorrhage
  • Abdominal pain
  • Vomiting
  • Abdominal distention
  • Tenderness
  • Dyspepsia
  • Dysphagia
  • Flatulence
  • Intestinal obstruction
  • Megacolon
  • Anorexia
  • Dysgeusia
  • Constipation
  • Increased ALT
  • Anaemia
  • Neutropenia

Precautions

Extra caution should be given in people with the following medical conditions:

  • Severe C. difficile infection
  • Inflammatory bowel disease (e.g. ulcerative colitis or Crohn’s disease, toxic megacolon)
  • Known macrolide allergy
  • Systemic infections
  • Moderate to severe liver impairment
  • Severe renal impairment
  • Pregnancy and lactation

Interactions

Fidaxomicin may interact with the following medicines:

  • Sodium picosulfate
  • Lactobacillus
  • Estriol
  • Mizolastine
  • P-gp inhibitor (e.g. ciclosporin)

If you have used Fidaxomicin more than the actual recommended dose, seek medical help immediately.
