Ergotamine is a partial serotonin agonist with vasoconstrictor effects. It is an ergot alkaloid and has an important oxytocic role on the uterus. It encourages the vasoconstriction of peripheral smooth muscles and cranial blood vessels, and slows down the central vasomotor activity.
Caffeine blocks the phosphodiesterase enzyme that causes an elevation of intracellular cyclic 3,5 adenosine monophosphate levels. It inhibits the peripheral pronociceptive property of adenosine and stimulates the central noradrenergic pathway which comprises an endogenous pain suppression system that causes its analgesic action. It raises the ergotamine enteral absorption and its vasoconstrictive activity. It also impedes the central adenosine receptors hence restoring the CNS and resp center, generating wakefulness, enhanced mental property, raises rate and respiration depth.
