DULOXETINE HCI

Duloxetine is a powerful inhibitor of neuronal reuptake of serotonin, norepinephrine and to a lesser degree dopamine. It has no affinity for adrenergic, muscarinic, cholinergic, histaminergic, narcotic, glutamate and GABA receptors. It additionally does not hinder the activity of monoamine oxidase.

Uses

Duloxetine HCI is used for the following conditions:

Warning

Do not give Duloxetine HCI to the following:

  • Hepatic impairment
  • Severe renal impairment
  • Uncontrolled hypertensionor narrow-angle glaucoma
  • Concomitant use with monoamine oxidase inhibitors(MAOIs) or within 14 days of discontinuing the MAOIs
  • Use withlinezolid or IV methylene blue
  • Concomitant use withpotent CYP1A2 inhibitors (e.g. ciprofloxacin, enoxacin)

  • Changes in behaviour
  • Suicidal ideation
  • Hypertension
  • Headache
  • Somnolence
  • Fatigue
  • Dizziness
  • Insomnia
  • Agitation
  • Tremor
  • Anxiety
  • Insomnia
  • Lethargy
  • Abnormal dreams
  • Nausea
  • Xerostomia
  • Palpitation
  • Hyperhydrosis
  • Decreased libido
  • Hot flushes
  • Sexual dysfunction
  • Vomiting
  • Constipation
  • Diarrhoea
  • Dyspepsia
  • Abdominal pain
  • Weight gain
  • Decreased appetite
  • Erectile dysfunction
  • Increased hepatic enzymes
  • Muscle spasms
  • Paraesthesia
  • Tinnitus
  • Blurred vision
  • Mydriasis
  • Nasopharyngitis
  • Hyponatraemia
  • Stevens-Johnson syndrome
  • Erythema multiforme
  • Hepatic failure with or without jaundice

Precautions

Precautions

Take extra caution in patients with the following medical conditions:

  • Hypertension
  • Gastroparesis
  • Mania or hypomania
  • Increased intraocular pressure or at risk of acute narrow-angle glaucoma
  • Seizure
  • Bleeding disorders
  • Mild to moderate renal impairment
  • Smokers
  • Pregnancy and lactation

 

Interactions

Duloxetine HCI may interact with the following medicines:

  • Tricyclic antideprssants (TCA)
  • SSRI, SNRI, lithium
  • Aspirin, NSAIDs, warfarin and other anticoagulants
  • Monoamine oxidase inhibitors (MAOIs)
  • Linezolid
  • Methylene blue
  • Potent CYP1A2 inhibitors (e.g. ciprofloxacin, enoxacin)
  • Overdose & Brands
  • Overdose symptoms include
  • Somnolence
  • Coma
  • Serotonin syndrome
  • Seizures
  • Vomiting
  • Tachycardia
  • Seek medical help immediately.

Overdose & Brands

If you think you have used Duloxetine HCI more than the usual dosage, please seek medical help immediately.
