Doravirine is a pyridinone non-nucleoside reverse transcriptase inhibitor (NNRTI) that reduces HIV-1 replication by competing with reverse transcriptase’s viral polymerase activity (RT). It interacts to a hydrophobic pocket in the p66 subunit of the RT enzyme’s p66/p51 heterodimer, producing conformational changes in the active site residues. This results in an inactive form, which prevents the chemical action of a polymerisation process from taking place.
