Acalabrutinib is a potent and irreversible Bruton Tyrosine Kinase Inhibitor (BTK). It inhibits BTK, an integral component of the pathways of the B-cell and cytokine receptor, which signals activation of the pathways necessary for B-cell proliferation trafficking, chemotaxis, and adhesion. By building a covalent bond with cysteine residue at the active BTK site to block BTK enzyme activity, acalabrutinib and its active metabolite (ACP-5862) reduce malignant B-cell proliferation and tumor development.
